31% Reduction in Senescence Biomarkers in 48 Hours
A replicative senescence model was developed using human fibroblasts from a 29-year-old donor. Senescence was induced by successive replication in culture up to passage 9. Fibroblasts were stimulated or not with Chronoglow™ at 0.2% for 48 hours, and senescence-associated β-galactosidase activity was measured. Cellular morphology was also analyzed via microscopic observation after vimentin immunostaining, as senescence significantly alters fibroblast structure. Chronoglow™ reduces SA-β galactosidase activity by 31% and restores a morphology closer to young fibroblasts.

Antioxidant evaluation was performed using an AmplexRed assay kit to evaluate ROS scavenging properties of Chronoglow™ at increasing concentrations. Hydrogen peroxide was used to induce oxidative stress and generate ROS, which were quantified via fluorescence. Chronoglow™ demonstrates dose-dependent antioxidant efficacy, with protection levels up to 90% at 0.2%.
